The SH2-containing protein tyrosine phosphatases References have been implicated as important regulators of signal DeFranco, A., and Law, D. (1995). Tyrosine phosphatases and the transduction (DeFranco and Law, 1995; Imboden and antibody response. Science 268, 263–264. Koretsky, 1995). Currently, there are two mammalian Imboden, J.B., and Koretsky, G.A. (1995). Switching off signals. Curr. members to this PTPase subfamily. They have a comBiol. 5, 727–729. mon structure consisting of two amino-terminal SH2 domains followed by a catalytic domain and a short tail. Owing to the independent isolation by several laboratories of the cDNAs encoding these phosphatases, a wide variety of names have been used in the literature. To facilitate thedissemination of informationconcerning these phosphatases, as investigators that have cloned cDNAs for mammalian SH2-containing phosphatase, we propose the name changes that follow. SHP-1 GenBank accession numbers are M68902, M77273, M90388, X62055; HGMW designation is PTPN6. SHP-1 has been previously known as SHPTP-1, SHP, HCP, and PTP1C. SHP-2 GenBank accession numbers are D13540, L03535, L07527, X70766, LO8663; HGMW designation is PTPN11. SHP-2 has been previously known as SHPTP-2, SHPTP-3, syp, PTP2C, and PTP1D. We would like to recommend that the names SHP-1 and SHP-2 be used in future publications.

The roles of EPIYA sequence to perturb the cellular signaling pathways and cancer risk
Abstract It was shown that several pathogenic bacterial effector proteins contain the Glu-Pro-Ile-Tyr-Ala (EPIYA) or a similar sequence. These bacterial EPIYA effectors are delivered into host cell via type III or IV secretion system, where they undergo tyrosine phosphorylation at the EPIYA sequences, which triggers interaction with multiple host cell SH2 domain-containing proteins and thereby...
متن کاملA Drosophila SH2-SH3 adaptor protein implicated in coupling the sevenless tyrosine kinase to an activator of Ras guanine nucleotide exchange, Sos.
A Drosophila gene (drk) encodes a widely expressed protein with a single SH2 domain and two flanking SH3 domains, which is homologous to the Sem-5 protein of C. elegans and mammalian GRB2. Genetic analysis suggests that drk function is essential for signaling by the sevenless receptor tyrosine kinase. Drk biological activity correlates with binding of its SH2 domain to activated receptor tyrosi...
متن کاملProbing the phosphopeptide specificities of protein tyrosine phosphatases, SH2 and PTB domains with combinatorial library methods.
Protein tyrosine phosphatases, SH2 and PTB domains are crucial elements for cellular signal transduction and regulation. Much effort has been directed towards elucidating their specificity in the past decade using a variety of approaches. Combinatorial library methods have contributed significantly to the understanding of substrate and ligand specificity of phosphoprotein recognizing domains. T...
متن کاملAPS, an adapter protein with a PH and SH2 domain, is a substrate for the insulin receptor kinase.
APS (adapter protein with a PH and SH2 domain) is the newest member of a family of tyrosine kinase adapter proteins including SH2-B and Lnk. We previously identified SH2-B as an insulin-receptor-binding protein and substrate [Kotani, Wilden and Pillay (1998) Biochem J. 335, 103-109]. Here we show that APS interacts with the insulin receptor kinase activation loop through its SH2 domain and insu...
متن کاملSpecificity of the SH2 domains of SHP-1 in the interaction with the immunoreceptor tyrosine-based inhibitory motif-bearing receptor gp49B.
Inhibitory receptors on hemopoietic cells critically regulate cellular function. Despite their expression on a variety of cell types, these inhibitory receptors signal through a common mechanism involving tyrosine phosphorylation of the immunoreceptor tyrosine-based inhibitory motif (ITIM), which engages Src homology 2 (SH2) domain-containing cytoplasmic tyrosine or inositol phosphatases. In th...
